Exolabs coliform medium contains a sugar linked to a dye which, when acted on by the enzyme galactosidase(produced by coliform bacteria including e.coli), turns the coliform colony a PINK color. Similarly, Exolabs coliform medium contains a second sugar linked to a different dye which produces a blueish-green color when acted upon by the enzyme glucuronidase. Because e.coli exclusively produces BOTH enzymes when present, e.coli bacteria can be identified by their PURPLE color(blue + pink).

Total coliform colonies= Pink.
Total e.coli colonies= Purple.
DISREGARD any light blue, blue-green, or white/yellow colonies.

For an accurate report, count all Purple colonies after 24 hours:

1. Since you sampled 5ml of water, multiply the number of e.coli colonies found times 20, to give you the # of colonies per 100ml of water.
EXAMPLE: 3 colonies found x 20 = 60 e.coli per 100ml of water.

2. EPA regulations for treated drinking water from public systems are 0 e.coli and other coliform bacteria per 100ml. Safe bathing and swimming water limits are approx. 100 e.coli per 100ml. Polluted harbors such as Hong Kong may register 7000 e.coli per 100ml.

1. Colorless/White/Yellow colony: No coliform or e.coli present.

2. Teal Green colony: Some glucuronidase reaction. No coliform or e.coli present.

3. Dark Green colony: Substantial glucuronidase reaction. No coliform or e.coli present.

4. Purple colony: Indicates presence of e.coli.

5. Pink colony: Some galactosidase reaction. Indicates coliforms, but not e.coli.

6. Dark Pink colony: Substantial galactosidase reaction. Indicates coliforms, but not e.coli.
